Defining the optimal operating conditions and configuration of a single-pass tangential flow filtration (SPTFF) system via CFD modelling

Single-pass tangential flow filtration (SPTFF) is used in biopharmaceutical manufacturing to reduce volumes and concentrate products. The staging of multiple membrane cassettes makes it possible achieve high concentration factors one pass, but at the same time increases system’s complexity. This work aims provide comprehensive understanding SPTFF both cassette level equipment by evaluating impact different operating conditions filter designs on performance. An experimentally validated computational fluid dynamics (CFD) model a commercial ultrafiltration was developed first study volumetric factor (VCF). results show that feed flux retentate pressure have substantial VCF. At low pressures (0.0–0.4 bar) VCF 1.2–2.9 achieved for fluxes 137–394 L m−2 h−1, while (0.6–1.0 up 10.5 achieved. subsequently applied concentration-dependent effects spatial distribution relevant physical properties. simulation help select suitable evaluate design with respect product accumulation.
